Jonas Salk menemukan vaksin polio tahun 1954 yang merupakan inactivated vaksin (berasal dari virus hidup polio yang dimatikan). Saat itu vaksin diberikan melalui suntikan. Pembuatan vaksin polio lalu dilanjutkan oleh Albert Sabin yang menghasilkan vaksin polio oral (diteteskan di mulut) yang kita kenal saat ini.
Dua tahun sebelum vaksin polio buatan Jonas Salk tersedia secara luas, jumlah anak-anak yang menderita polio sangat banyak. Di AS saja jumlahnya lebih dari 45.000 anak. Tapi di tahun 1962 jumlahnya berhasil turun menjadi 910.
Jonas Salk merupakan dokter lulusan New York University School of Medicine tahun 1993. Ia lalu bekerja sebagai staf dokter di Mount Sinai Hospital di New York.
Ia kemudian mengikuti bakatnya sebagai peneliti di Universitas Michigan di mana ia bekerja mengembangkan vaksin flu atas permintaan tentara AS. Di tahun 1947, ia ditunjuk menjadi direktur laboratorium riset virus di University Pittsburgh School of Medicine. Di tempat ini ia kemudian mengembangkan teknik yang membantunya menemukan vaksin polio.
Jonas Salk mengerjakan kembali ide yang sudah ada tentang vaksin menggunakan kekebalan yang dihasilkan tubuh dengan menggunakan virus yang tidak aktif. Ia lalu mendapatkan dana untuk mengembangkan vaksin polio.
Vaksin yang dihasilkan tersebut pertama kali diuji coba pada monyet baru kemudian ke pasien anak cacat yang sudah menderita polio. Kemudian dicoba juga ke sukarelawan, termasuk staf laboratorium, termasuk Jonas Salk, istri dan anaknya. Ternyata tidak ada efek samping negatif yang mereka rasakan.
Tahun 1954 dilakukan uji coba vaksin secara nasional pada satu juta anak berusia 6-9 tahun yang kemudian dikenal sebagai Pionir Polio. Setengah anak mendapat vaksin dan sisanya diberi plasebo. Pada 12 April 1955, vaksin tersebut dinyatakan aman dan efektif.
Jonas Salk memilih untuk tidak mematenkan vaksin tersebut dan tidak mendapatkan imbalan uang untuk penemuannya tersebut. Ia lebih suka jika vaksin tersebut didistribusikan seluas-luasnya. Salk meninggal dunia di usia 80 tahun pada 23 Juni 1955.
